Liver Extract Solid

No.

Instruction

Initials
Treat the liver powder with acidified alcohol
Cover & stir for 30 minutes
Filter via vacuum and Buchner
Treat solid residue with 50% ethyl alcohol
Cover & stir for 30 minutes
Filter via vacuum and Buchner
Combine the two filtrates and Discard residue
Filter the mixture to clarify (yield approximately 1000ml for 200Gm of liver powder)
Evaporate the mixture in hood with heated air source (loose approximately 80% of the starting volume)
Prepare a number of tarred filter papers (mark w pencil)
Treat the concentrate with anhydrous or 95% ethyl alcohol.  Do in small amounts.  Swirl for 30 - 60 seconds.  Filter this carmel-colored suspension through weighed paper.  Continue using the same piece of paper until it seems fully loaded/clogged.
Dry the filter papers
The new filtrate may be concentrated again but each time the yield will be less.  It is probably not worth the effort to repeat the concentration/filtering process.
Weigh the dried filter papers
Subtract the weight of the blank filter papers from the weight of the filter paper that has material on it.  The difference is the amount of liver extract generated.
Cut the filter paper into small pieces (use clean scissors)
Place these small pieces in a beaker
The final concentration is to be 20mg/ml (sterile).  So. use the weight determined above to calculate the final volume to be made.  Add 80% of this volume to the shards of paper.
The active liver extract will dissolve off the paper
Filter via vacuum and Buchner
Filter through 0.8 micron filter and POSSIBLY a 0.45 micron
Filter through 0.22 micron to sterilize
Bring to final volume with SWFI
